POS1441 OCULAR AND PERIPHERAL BLOOD IMMUNE PHENOTYPES SUGGEST TRANSMIGRATION OF CD16+ MONOCYTES TO AN IMPORTANT EFFECTOR SITE IN BEHÇET’S DISEASE

چکیده
Background Cellular immunity of Behçet’s Disease (BD) remains poorly understood. Previous work has provided clues pointing to most innate and adaptive immune cell types in BD, but strong signals from non-immunogenetic studies are rare often inconclusive. Objectives Here we aimed identify BD/HD discriminant single profiles semi-biased targeted approaches determine their significance at a BD relevant effector site. Methods We utilized multi-parametric flow cytometry dissect cellular phenotypes PBMC untreated patients (n=27) HD (n=22) consisting predominantly active ocular major vascular subjects. Data were subjected supervised machine learning (CITRUS) results verified with gating. also analyzed anterior chamber (AC) fluid cells autologous uveitis subjects scRNA seq. Results CITRUS identified CD16+, CD14low, CD4low, CD3-, CD19- as distinguishing expression pattern an FDR >0.05. Targeted gating confirmed highly significant differences large effect sizes vs for “non-classical” (CD14lowCD16hi) “intermediate” (CD14+CD16+) monocytes decreased frequencies compared peripheral blood. “Classical” (CD14++CD16-) more abundant than HD. CD16+ dendritic (DC) significantly PBMC. CD14+ showed high abundance the AC during co-expressed CD16 far frequently Conclusion Significantly lower monocyte DC subsets strongly point importance BD. The co-expression eye relative frequency blood, suggests transmigration or post-migrational interconversion within rather stochastic process. REFERENCES: NIL. CD16+ monocytes control T-cell subset development in immune thrombocytopenia.
Immune thrombocytopenia (ITP) results from decreased platelet production and accelerated platelet destruction. Impaired CD4(+) regulatory T-cell (Treg) compartment and skewed Th1 and possibly Th17 responses have been described in ITP patients. The trigger for aberrant T-cell polarization remains unknown. Because monocytes have a critical role in development and polarization of T-cell subsets, w...
متن کاملIncreased subpopulations of CD16(+) and CD56(+) blood monocytes in patients with active Crohn's disease.
BACKGROUND Circulating monocytes may be subdivided according to the presence or absence of the Fcgamma receptor CD16 and the neural cell adhesion molecule CD56. Monocytes classified into these subpopulations are characterized by distinct phenotypic and functional features. We hypothesized that patients with active Crohn's disease differ in their peripheral monocyte subpopulations. METHODS Usi...
متن کاملPivotal Role for CD16+ Monocytes in Immune Surveillance of the Central Nervous System.
Monocytes represent a heterogeneous population of primary immune effector cells. At least three different subsets can be distinguished based on expression of the low-affinity FcγRIII: CD14(++)CD16 -: classical monocytes, CD14(++)CD16(+) intermediate monocytes, and CD14(+)CD16 ++: non-classical monocytes. Whereas CD16 -: classical monocytes are considered key players in multiple sclerosis (MS), ...
متن کاملdevelopment and implementation of an optimized control strategy for induction machine in an electric vehicle
in the area of automotive engineering there is a tendency to more electrification of power train. in this work control of an induction machine for the application of electric vehicle is investigated. through the changing operating point of the machine, adapting the rotor magnetization current seems to be useful to increase the machines efficiency. in the literature there are many approaches wh...

Our aim was to characterize glomerular monocytes (Mo) infiltration and to correlate them with peripheral circulating Mo subsets and severity of lupus nephritis (LN). Methods. We evaluated 48 LN biopsy samples from a referral hospital. Recognition of Mo cells was done using microscopic view and immunohistochemistry stain with CD14 and CD16. Based on the number of cells, we classified LN samples ...
